Well, if you've seen The Incredible Hulk (2008), Iron Man, and Iron Man 2, and watched past the credits of all those films, you'll truly appreciate Thor. Be sure to watch past the credits, though. They throw in so many references and cameos in the film and I really appreciated it. :P

I haven't actually read the comics, but I love the movies.
